چکیده

In order to avoid the problem of an uncoupling effect in stimulation of potential Hill reaction activity during extraction with heptane. Hill reaction activity was measured with uncoupled spinach chloroplasts. The reactions were also run under aerobic conditions. Under these conditions a correction for a 3-(p-chlorophenyl)-1, 1-dimethylurea insensitive, photostimulated O(2) uptake reaction is necessary, especially with extracted chloroplasts. The reaction displayed proper Hill reaction stoichiometry in untreated and restored systems if these corrections were made. Plastoquinone A always restored some activity to the extracted chloroplasts but never restored activity to the original level. Additional activity could be restored by the crude extract. It appears that there is a clear requirement for plastoquinone A in the Hill reaction but the restoration of maximum activity depends upon additional factors. The variability of response to plastoquinone A which has been reported in various laboratories may be determined by the degree of extraction of the other materials.

The specificity of plastoquinone as a cofactor for photophosphorylation.

The requirement for plastoquinone in the photosynthetic phosphorylation activity of spinach chloroplasts has been indicated in an earlier publication (1). Removal of endogenous plastoquinone from lyophilized chloroplasts by heptane extraction caused a loss of photophosphorylation activity. The photophosphorylation activity could be partially restored by addition of pure plastoquinone to the ext...

Studies on Plastoquinonez-1. Determination of the Concentration and Oxidation-reduction State of Plastoquinone in Isolated Chloroplasts

It was shown by Cranes that plastoquinone is localized in the chloroplasts of higher plants, and there is now a good deal of evidence which suggests that it bctions as an oxidation-reduction carrier in the photosynthetic electron transport system. Bishop’ showed that when lyophilized chloroplasts were extracted with organic solvents they lost their Hill-reaction activity, but that this could be...

Electron transport in chloroplasts. I. A combined requirement for plastoquinones A and C for photoreduction of 2,6-dichloroindophenol.

Long term extraction of lyophilized chloroplasts with heptane removes plastoquinones A 45 and C. Hill reaction activity as measured by photoreduction of 2,6-dichloroindophenol is decreased in proportion to the extraction of plastoquinone C, whereas up to 40% of the plastoquinone A can be removed without affecting photoreductase activity. Some restoration of activity may be obtained by adding pl...

Site of Action of 2,5-Dimethoxy-3,6-Dichloro-/?-Benzoquinone in the Photosynthetic Electron Transport Chain

Dichlorodimethoxy-/?-benzoquinone (DCDMQ) was tested for its site of action in the photo synthetic electron transport chain. Hill reaction mediated by DCDMQ was insensitive to DBMIB (1 nm) but sensitive to DCMU, suggesting its site of action before plastoquinone but after Q the primary electron acceptor of photosystem II. Extraction of freeze-dried chloroplasts with heptane and analyzing their...
